Harding-Passey melanoma: Description, Causes and Risk Factors:Harding-Passey melanomaThis tumor originally found in brown mice; now used as inducer of nonmetastatic melanoma when transplanted into other mice.Harding-Passey melanoma is one of the best tumors for producing melanin and is used extensively to study biosynthetic melanin pathways and their regulations in mammals, since melanocytes are difficult to obtain from normal skin.Pigment cells are derived from the neural crest and are included in the paraneuron group maintaining some common functional characteristics. Thus, the Harding-Passey melanoma can be a good model to approach the energetic requirements that allow us to compare melanomas with other perineuronal and tumor tissues.A high rate of anaerobic glycolysis has been described in most tumor tissues and many explanations have been proposed. Recently, insulin receptors have been described in tumor cells but the effect of this hormone on glucose transporters at the plasma membrane has not been discussed.Overall glucose utilization by Harding-Passy melanoma cells were less than in other tumor cells, such as Ascites tumor and neuroblastoma and also when compared to normal neural tissue, either in vivo or in isolated cells.Studies shown that the Harding-Passey melanoma is heat-sensitive. Its heat-sensitivity increases as the tumor develops in the host. Excision achieved a higher percentage of long-term cure than hyperthermia, during the time period of tumor development studied in these experiments, but did not give the host long-term protection from subsequent tumor challenge.Symptoms:There are no known symptoms.Diagnosis:In some cases electron microscopic observation of Harding-Passey melanoma revealed that the melanoma consisted of a melanin producing cell, a melanin containing cell and a melanin forming and containing cell. Melanosomes were present in the melanin containing cell in the form of a melanosome complex which is now thought to be some sort of a lysosome formed by autophagy. The melanin producing and containing cell is assumed to be the aged melanocyte and the melanin containing cell to be the older melanocyte.Treatment:The successful treatment of melanoma is a challenge to oncology since its high metastasis rate and evolution is usually diagnosed at stages of development when its surgical remission is ineffective.There is no known treatment. Harding-Passey melanoma homogenates with UV and by doing so induce melanin formation within melanosomes which imparted on them a greater density and thus made it possible to easily separate them from mitochondria by density gradient centrifugation.NOTE: The above information is educational purpose.
